Staining for MAP-2 revealed that neurons are not affected by their proximity to the wire, as both neuronal soma and processes maintained in the same density next to the wire as in areas of culture away from the foreign body (Figure 4A). To verify whether the neurons in culture are healthy and electrically active, patch clamp recordings on individual neurons were performed. Neurons identified by morphological markers and patch clamping were performed on neuron-glia culture preparations, at different post treatment time points, while the cells were in their medium treatment. Action potentials were clearly distinguished (Figure 4C) and had a physiologically accurate shape profile (Figure 4D). Finally it was confirmed that the neurons would respond to stimulation such as depolarizing current by electrically activating the cells into producing action potentials (Figure 4E).

According to Clontech’s informations, each total RNA sample was extracted using a modified guanidine thiocyanate method with quality control tests which guarantee the presence of full-length transcripts, rare transcripts and no genomic DNA. Each sample corresponds to a pool of total RNA extracted from several normal human tissues from the same localization.
